Effects of Methoxyflurane Anesthesia on Ultrastructure of Rat Kidney

Abstract
The author studied the uItrastructural changes of rat kidney follo~.vin~ inhalation anesthesia with r.:ethoxyfiurane, utilizin,; halothane for control. Fifteen all;ino rats of both see:es, weighin~-bet~.t-een 200 and 250 gm., were used in this study. 1 he rais were divided into three groups; three as normal control, six as control with halothane anesthesia in the moderate depth for two hours, and the rest six were anesthesized with metho~yflurar.c in fire a~odcratc deT;th for two hours.
The results were st;mmarized as follow:

EIectron microscopically, in the rats immediately after ha¢¥othane anesthesia, mild proliferation of sirD and slight degeneration of mitochondria were obsen-e3 in the tu¢¥uular epit¢¥_zelial ells of the cortex and the medulla. In the 24 hours after halothane anesthesia, slight degeneration and diminution of mitochondria, moderate proliferation of shR, appearance of Iysosomes in varying shape and focal thickening of basement membrane were observed in the proximal tubules. These changes were more prominent in the tubules of the mec[ulia than in t¢¥7e cortex. In the rats immediately after methoxyflurane anesthesia, mild proliferation of sEr was observed in the prosirnai tubules, and mild degeneration of mitochondria and slight increase of lysosome~ were observed in tlae tubules of the medulla. In the 24 hours after methoxyflirane anesthesia, widening of the tubular lumen, swelling and degeneration of r.;itachondria, increase of varying shaped lysesemes, appearance of focal cytoplasmic degradation and mild thickening of basement membrare were observed in the proximal tubules. In the to ales of the medulla; market¢¥ proliferation. c sER, increase of lysosomes, diminution of mitochoncria, focal thickening of basement membrane and marked diminution of basal intussusceotion were observed.
The results obtained in this study suggest that methoxyflurane anesthesia causes focal thickening of basement membrane, diminution of basal intussusceptic;n, and mild degenerative changes in the tut~ules of the cortex and the medulla. These changes ~~re considered to be related with renal insufficiency induced by methoxyflurane anesthesia in human practice.
